Sažetak
A virus was isolated from the rose with symptoms of linear mosaic and ring spot on the leaves, found in the surrounding of Zagreb. On the basis of herbaceous plant reaction "Shirofugen" test as well as serological investigation, it was assumed that the cause of this disease is Prunus necrotic ring spot (PNRV).The isolate R1 obtained from rose reacted positively with the serum against serotype G of PNRV. This virus was also found on peach cultivar Dixired in environs of Zagreb. The identification of the virus again based on reaction of herbaceous host and woody indicator Prunus serrulata (Shirofugen). With the isolate B obtained from and peach we performed we performed two successful purification using the HEPES-buffer in the first trial and HEPES-buffer and PEG-600 in the second one. When isolate B was conserved in dry leaves according to the method of McKinney it remained infectious for two full years. However, the isolate R lost its infectivity after eight months. A large number of peach trees grown in the areas of Zadar (Bastica) and Zagreb were tested serologically on the presence of PNRV using as a source of antigen the leaf buds. During these investigations double diffusion in agar-gel and latex method were applied. In these experiments it was established that 67 % of peach trees were infected with PNRV. These methods can be used beneficially in the selection of healthy parent plants.
