Sažetak
Peptidoglycan monomer (PGM ; dissacharide-pentapoptide) iolated from Brevibacterium, is an effective immunolomodulator and antitumor agent in mice. One intravenous injection (6o mg/kg) of PGM, administrated when the tumor was about 3 mm, significantly reduced the growth of an implanted fibrosarcoma (FSa, methylcholanthreneinsduced). In this investrigation the effect of the injection om in vivo phagocytosis by resident peritoneal macrophages and on tumor-specific immunity (footpad test) was studied. Mice of the CBA/H/Zgr strain, kept under conventional conditions, were used. Phagocytosis was examined 8, 16, 24, 48 or 72 hr after the PGM injection. Cells from the nonirritated peritoneal cavity of injected mice were coincubated with yeast cells (, Saccharomyces cerevisiae) for 10 minutes, and the percent of macrophages with ingested YC (phase-contrast microscope) was determined. PGM stimulated the phagocytosis whichwas most expressed 24 hr after the injection. To test the specific antitumor immunity, PGM was injected two or four days after injection of FSa cella and the footpad reaction was scored on the 8th day. The injection of PGM has not increased the cellular immunity to FSa. It may concluded that the single injection of PGM stimulated phagocytosis by peritoneal macrophages, but did not stimulate specific antitumor immunity in injected mice. Thus, the antitumor effect of the single PGM injection could result primarily from PGM's potentu+ial to activate macrophagrs. 8YC, Saxxharonyces cerevisiae 9
