Sažetak
Following Commissions Implementing Decision (2014/87/EU) from February 2014, an official annual survey for the presence of the specified organism on plants and plant products has been undertake along the coastal territory as a protective preventive measure against the introduction of harmful organism Xylella fastidiosa (Well and Raju). Since the main entry pathway for the specified organism is the movement of plants for planting, based on the data on imports of plant material from the past few years, our work has been focused to the young plants of Nerium oleander L. which is present in all urban and semi urban zones, by the roads and highways, same as plants of Olea europaea L. used for horticultural landscaping in urban zones and backyards. Beside survey in urban and semi urban area, survey in intensive and traditional olive plantations in regions of Dalmatia and Istria has been undertaken. Survey was organised in six counties along the whole coastal area in collaboration between Institute for Plant Protection (PPI) and Regional Phytosanitary Inspection Units. Survey was done by visual inspections of around forty towns and cities that are tourist-oriented, characterized by high traffic and patchy horticultural decorating using principal host plants of targeted harmful organism. Main host plants and other potential host plants were visually inspected in order to find plants showing symptoms of leaf scorching, dying or decline. Such samples were collected for serological and molecular assays conducted by the Laboratory for bacteriology, Institute for Plant Protection. A total number of eighty samples of oleander and olives were analysed. As a first method of detection ELISA was used. To confirm the results of the first method of testing, an polymerase chain reaction (PCR) with primers for amplification of the 16S rRNA gene and gyrB genes were used. Results of the first year of survey in Croatia shows that harmful organism Xylella fastidiosa (Well and Raju) was not found after extensive visual inspections and laboratory assays.
