Sažetak
Cytotoxic T lymphocytes and natural killer (NK) cells represent two major types of immune cytotoxic cells that have an important role in immune response eliminating pathogen-infected cells. Both of these cells have a possibility to kill by two distinct signal transduction pathways. The first is the granule exocytosis pathway mediated by direct releasing of cytotoxic granules (perforin and granzymes) into the synapse between the effector and target cell. Perforin and granzymes are stored in the cytoplasmic granules of the cytotoxic cells. The aim of this study was to examine alteration of cell mediate immunity and cytolytic molecule perforin in different lymphocyte subpopulation in peripheral blood of subjects situated near gasoline industry compared with healthy laboratory donors.Venous blood samples were taken from all examinees and peripheral blood leukocytes (PBL) were isolated. Phenotype of lymphocytes and perforin expression were analyzed using intracellular and surface immunofluoresecncy and flow cytometry (FACSCalibur). Statistical analysis was done in computer program Statistica 7.1 (StatSoft, Inc., Tulsa, OK, USA). Graphs and tables were done in computer program Microsoft Excel.The results demonstrated that 17% of PBL in all examine group are perforin positive (total perforin is significantly lower in exposure than in control group). Percentages of double positive cells showed significant decrease in all examine lymphocyte subpopulations (CD3+ P+, CD4+P+, CD8+P+, CD16+P+ and CD56+P+ cells) compared to controls.Our data suggest that human perforin is differentially expressed in distinct sub- populations of peripheral blood lymphocytes of people exposure to gasoline and diesel, implicating the important role of perforin positive cells in naïve immune response and cytotoxic activity of peripheral blood lymphocytes.
