Sažetak
Objective: Thyroxine (T4) is the principal prohormone produced by follicular cells of the thyroid gland. It is essential for regulation of metabolic processes throughout the body. T4 in the circulation is derived from thyroidal secretion and becomes activated upon its conversion to triiodothyronine (T3). Local activation of prohormone T4 at the tissue level is increasingly recognized as an important mechanism of regulation of thyroid hormone action. The aim of this study is to identify genetic loci associated with serum level of T4. Methods: Our genome-wide data contained 1012 healthy subjects from Split region. For the purpose of this analysis, we excluded individuals taking thyroid medications. After applying exclusion criteria, the genome-wide association analysis of T4 on 966 individuals was performed accounting for genetic relatedness among individuals. Associations with T4 serum levels were assessed by linear regression using residuals adjusted for gender and age. Results: Our study showed a novel genome-wide significant association of T4 levels on chromosome 12 at rs150958959 (β=1, 45, SE=0, 24, p=1, 81x10-09, MAF=0, 02). The identified locus is located on chromosome 12 near FLJ37505 gene. Conclusion: We identified novel locus associated with T4 levels in the general population from Split region. Given the low minor allele frequency of the identified SNP, this finding needs further confirmation in a larger sample.
