Sažetak
Lactobacillus amylovorus is one of a number of lactobacilli which possess amylolytic activity and thus is able to exploit starch directly to form lactic acid. Starch is a polymeric carbohydrate consisting of a large number of glucose units joined by α-glycosidic bonds. Lactobacillus amylovorus produces extracellular α-amylase which can degrade starch and can be used to produce lactic acid directly from starch without additional processes like liquefication and saccharification. Due to these characteristics, it can be used as a starter for developing an economically viable process for lactic acid production from cheap materials containing starch. The main goal of this research was to identify the gene encoding an interesting α-amylase. For the purpose of identifying amylase gene, PCR reactions and preparation of genomic library were conducted. Certain results have shown that the sourced strain has not been correctly identified, and in order to continue the project, a synthetic amylase gene has been ordered, based on a recently published genome sequence of Lactobacillus amylovorous DSM 20531.
