Sažetak
Leuconostoc mesenteroides belongs to the group of industrially relevant biocatalysts and its modern implementation in a manufacturing of whole range of mass products is far beyond acidification and flavouring of certain feedstock. Genetic potential of this mesophylic heterofermentative lactic acid bacterium comprises functional gene(s) for sucrose phosphporylase (EC 2.4.1.7), very well characterized enzyme used for glucosylation reactions in the industrial scale. In this work we investigated employment of permeabilized cells of L. mesenteroides in order to perform targeted glucosylation reaction in which sucrose served as an glucosyl donor while glycerol was a glucosyl acceptor. The bacterium was cultivated in the semi-defined medium during batch experiments that were carried out at 29±1C and nonmaintained pH value, and harvested cells where permeabilized by (i) ultrasound treatment (37 kHz, 90 W, 29±1℃ continuously over 4 hours and periodically two times over 30 minutes with 30 minutes pause inbetween the each treatment) and (ii) non- ionic detergent Tween-80 (0.4% and 0.6%). Possible formation of α-D-glucosylglycerol was indicated by a new peak that was eluted from ion-exchange column between sucrose and fructose. To estimate efficiency of performed transglucosylation reaction, separation, concentration and additional characterization of potential product is needed.
