Cytomegaloviruses (CMVs) induces extensive rearrangements of the cellular membrane system for assembly of CMV virions during the cytoplasmic phase. The rearrangement itinerary and mechanisms are still poorly characterized. In this study, we identified the stage of the endosomal route which is perturbed during the early phase of murine CMV (MCMV) infection. We demonstrated that MCMV infection does not affect endocytic uptake and early endocytic trafficking transferrin receptor (TfR), but arrests internalized TfRs is in enlarged endosomes near the juxtanuclear area together with internalized cell surface Major histocompatibility class I (MHC-I) proteins, mannose 6-phosphate receptor and epidermal growth factor receptor. TfR and MHC-I recycling is inhibited suggesting that MCMV infection affects early-endosomal maturation at the stage of sorting endosomes and cargo transport to the endosomal recycling compartment. An MCMV- encoded function that perturbs endosomal trafficking is expressed after three hours of MCMV early gene expression, and the first detectable functional consequences of endosomal perturbation are displayable after for hours of infection. Juxtanuclear concentration of early endosomes may represent the earliest event in the development of the MCMV assembly compartment which occurs very early in the infection.
