AIM: We aimed to assess the genotoxic potential of orthodontic adhesives on human lymphocytes. MATERIAL AND METHODS: Five adhesives were tested: Transbond XT ( 3M Unitek, USA) Transbond Plus (3M Unitek, USA), Light Bond (Reliance Orthodontic Products Inc., USA), Light Bond with Fluoride (Reliance Orthodontic Products Inc., USA) and Geristore (DenMat, USA). In a simulation of the clinical procedure, each adhesive was placed between two cellulose strips, covered with a metallic premolar bracket and illuminated for 10 or 20 seconds. Lymphocytes were obtained from a young, healthy, non-smoking donor. For the micronucleus assay, lymphocyte cultures were treated with samples for 24 h. Cell culture medium was used as negative and ethyl methanesulfonate in concentration 10 µg/mL of culture as positive control. Micronuclei were counted in 2000 binuclear cells per tested material. The experiment was performed in duplicate. Chi-square was used in statistical analysis. RESULTS: Differences in micronuclei formation between samples of the same adhesive with regards to curing time were not significant. Transbond Plus showed the highest frequency of micronuclei in both curing times, yet the difference was not significant. A significant increase in the formation of nuclear buds was also observed for Transbond Plus after 10 s of curing, however, it lacked biological relevance. CONCLUSION: Transbond Plus may induce increased DNA reparation, however, none of the tested adhesives showed genotoxic potential.
