Sažetak
Many viruses, including herpes simplex virus 1 and 2, have been found to express functional microRNAs. Viral miRNA can target both viral and cellular transcripts and can have a profound effect on virus replication. For example, HSV-1 expresses miRNAs that target important virus genes ICP0 or ICP4. However, although HSV-1 miRNAs have been discovered more than 10 years ago, little is known about their biogenesis and regulation. The main aim of this study is to identify a promoter for miR-H1, a virus encoded miRNA abundantly expressed during productive infection. Thus far, by using different bioinformatics tools we predicted the promoter region and the transcriptional termination site for the transcript that might give rise to miR-H1. To test if the predicted promoter region indeed contains the promoter we have generated a dual-luciferase reporter assay based on a commercially available system pmiR-GLO (Promega). We are planning to replace the original promoter (PGK) for the expression of a reporter luciferase with the 1.5 kb genomic region that spans the predicted promoter derived from the HSV-1 strain KOS. Next, we plan to test the activity of luciferase by introducing different mutations to identify the exact miR-H1 promoter sequence. Our current results and plan of our future experiments will be presented.
