Sažetak
Water deficit is one of the crucial factors causing environmental stress and it can be considering a particular climate region, either long-term or short-term and random. Sessile organisms like plants had to develop special strategies in order to overcome and survive periods of drought some of which vary depending on the genotype. This thesis involves exposure to drought of four different ecotypes of Arabidopsis thaliana native to different climate regions and establishing their relative tolerance to water deficit on the level of proteome. We compared peroxidase activities as indicator of oxidative stress and relative concentrations of Hsp70 (heatshock protein) as indicator of general stress. We also determined relative concentrations of intrinsically disordered proteins (IDPs) as they are involved in integration of various signalling pathways and therefore take part in many plant processes in general, including abiotic stress responses. Compared to control, a rise was shown in both peroxidase activity and Hsp70 expression during drought conditions. Expression of some IDPs also changed after exposure to drought in comparison to control. In addition, 25 IDPs were analyzed by mass spectrometry and bioinformatics tools have evaluated their sequence structure disorder. Twenty-one of them confirmed to have at least one long disordered region. Difference in protein expression were noticed among ecotypes.
