Sažetak
Problem Decidual lymphocytes (DL), containing mostly NK cells of unusual CD3-CD16-CD56bright+ phenotype, infiltrate human first trimester pregnancy decidua. These cells are immunocompetent and express high number of cytolytic molecule-perforin in their cytoplasm. Their cytolytic potential is strongly regulated by cytokine network operating in decidua. In this study we have investigated the effects of short term (18 hr) and long term (72 hr) stimulation by IL-12, IL-15 and IL-18 on cytolytic activity of DL against NK sensitive K-562 targets. Methods Decidual cells from the Lymphoprep interface were stimulated for 18 hr and 72 hr with IL-12 (50 pg/ml), IL-15 (5 ng/ml), IL-18 (10 ng/ml) or cultured in the medium only. Cytolytic activity of nonadherent cells-DL against NK sensitive K-562 cells was measured in 2 hr PKH-26 (red) cytotoxicity assay and analysed by flow cytometry. Results IL-12 increases cytolytic activity of DL against K-562 targets in a short term stimulation. IL-15 in a long term stimulation upregulates cytolytic activity of DL. On the contrary, IL-18 in a short stimulation only, increases cytolytic activity of DL. IL-12 or IL-15 combined with IL-18 in a short term stimulation do not change cytolytic activity of DL increased by IL-18. Conclusions IL-12 and IL-18 seem to be fast regulating factors of cytolytic activity of DL against K-562 targets. Contrary, IL-15 plays an important role in the regulation of cytolytic activity of DL in a long term stimulation.
