Natural killer (NK) and cytotoxic T lymphocytes (CTL) contain in their granules cytolytic molecule perforin (P). The aim was to analyse phenotypic profiles of P+ lymphocytes in the peripheral blood (PBL) of nonpregnant women, first trimester of pregnancy (FTP) and term pregnancy (TP) and FTP decidual lymphocytes (DL). Significant decrease of CD16+ cells in FTP-PBL was found and manifold increase in the time of parturition (TP), when total P+ cells increase as well. NK and CTL showed drastic changes both in subpopulations and during various phases of pregnancy. The analysis of the phenotype of P+ PBL showed significant increase of CD4+ cells in FTP and CD8+ and CD56+ in both FTP and TP groups, but surprinsigly drastic decrease of CD16+ cells from 45% in NP group to 4% in FTP group, and very high expression (67%) at the end of the pregnancy. The analysis of P-expression cells as a fraction of total PBL showed an increase of CD3+P+, CD4+P+ and CD56+P+ during pregnancy, but simultaneously drastic decrease of CD16+P+ cells in the first trimester (2%) and increase at the end of pregnancy (25%). The overlapping of CD3+P+ and CD56+P+ cells among P+ is suggesting that a part of CD3+ cells are CD3+CD56+P+. The phenotype of FTP decidual NK cells was CD3-CD56bright+Pbright+ but only 2% of them are CD16+. NK activity of FTP-DL against sensitive line K-562 is much lower than PBL and both can be only slightly upregulated by rIL-2 incubation. The CD16 molecule is the IgG FcRIII that is exclusively responsible for ADCC. It can be concluded that this mechanism of CMC is strongly suppressed in the FTP (downregulation of CD16 expression in CD3-CD56+ PBL and particularly in FTP-DL) but upregulated in the time of parturition.
