Sažetak
Introduction Hyperglycemia in diabetes causes a number of complications. Episodes of hyperglycemia after eating are particularly harmful and partial reduction of glucose uptake in small intestine might prevent or delay the development of complications. It was known that glucose, resulting from digestive process, goes across intestinal membrane together with two sodium ions by Na+-dependent glucose transporter (SGLT1). By using calcium Zeolite A (Ca Zeolite A) and its characteristic of binding Na ions inside the structure, we investigated the possibility of reducing glucose uptake. Material and methods Human colorectal adenocarcinoma (CaCo-2) cells were used as a model for intestinal glucose absorption. The cells were grown in completed DMEM medium or completed medium previously treated with Ca Zeolite A (10 mg/ml, stir 2h, room temperature). MTT test was performed to investigate toxicity of Ca Zeolite A on cells and colony forming assay (CFA) was done to detect proliferative capacity of the cells. SGLT-1 and ZO-1 proteins were detected by immunofluorescence. For detection of glucose uptake we used fluorescent derivate of D- glucose (2-[N-(7- nitrobenz-2-oxa-1, 3-diazol-4- yl) amino]-2-deoxy- d-glucose (2-NBDG). Results The viability of CaCo- 2 cells, after growing in medium pretreated with Ca Zeolite A, were about 80%, and their capacity of forming colony was slightly lower compared to non-treated cells. CaCo-2 cells expressed SGLT-1 not only on the membrane than also inside cells. ZO-1 protein, which is marker of cell-to-cell contact, is detected at the border between cells, what is characteristic of enterocyte.
