Sažetak
Idiopathic dilated cardiomyopathy (iDCM) is a primary myocardial disorder of unknown etiology characterized by reduced contractility and ventricular dilation involving the left or both ventricles (1). It is one of the most common heart diseases in dogs, with the prevalence of 0, 5% in general canine population (2). DCM occurs more frequently in large and giant breed dogs and rarely in medium, small or mixed breeds (3) and carries a poor prognosis with progression to heart failure. Traditionally, evaluation of heart function is performed by electrocardiography, radiography and echocardiography, which are time consuming and expensive. Serum biochemical markers, such as cardiac troponins and natriuretic peptides, are being used in human medicine in addition to these methods as a convenient tool for diagnosis and prognosis of heart disease. In the recent decade studies of biomarkers of cardiac diseases in dogs also showed promising results (4). The aim of the present study is to identify differences between serum biochemistry profiles of dogs with diagnosed iDCM (N=8) to those of healthy dogs (N=8). Each group consisted of 4 male and 4 female dogs of various breeds, with the average age of 7, 125 ±1, 642 in iDCM group and 8, 438 ±2, 718 in healthy group. Serum biochemistry parameters were measured using commercial kits in an automatic analyser (Olympus AU 600, Olympus Diagnostica GMBH). The statistical analysis was performed using GraphPad Prism software and the level of significance of p < 0.05 was considered statistically significant. Creatine kinase (CK), C reactive protein (CRP), lactate dehydrogenase (LDH), serum urea and triglycerides levels were significantly increased in serum samples of dogs with iDCM compared to healthy dogs, while magnesium levels were significantly lower. Both CK and LDH were previously reported as markers of myocardial injury and necrosis (5), while CRP is well recognized as a sensitive systemic marker of the acute phase response to most forms of tissue damage and inflammation in human and veterinary medicine (6). The next step of this study is to perform liquid chromatography tandem-mass spectrometry analysis of serum proteome using gel-free quantitative proteomics approach in order to get further insight into systemic status of dogs affected with iDCM and discover new diagnostic/prognostic biomarkers of this disease.
