Sažetak
The aim of this study was to evaluate the potential of using MRI relaxometry to characterize engineered cartilage tissue grafts used for treating articular cartilage defects in sheep. The MRI results were compared to standard histological methods. Cartilage tissue grafts were engineered from nasal chondrocytes seeded on Chondro-Gide scaffold. After the biopsy of nasal septum cartilage in sheep, specimens were dissected, and digested in a 0.15% collagenase solution. The isolated cells were proliferated in a proliferation medium for 12 days, and then seeded on a scaffold and cultured for either 2 (N-CAM group) or 14 days (N-TEC group). For MR measurements, the cultured grafts were placed in 2 ml cryotubes containing DMEM and scanned before and after Gd-DTPA2- based contrast agent (Magnevist®, Bayer Schering Pharma, Germany) was added. High resolution T1 scans (90x90x600 um3) were obtained using a 7 T preclinical MRI system (BioSpec 70/20, Bruker Biospin, Germany), and T1 maps were calculated from the measured data. After MR imaging specimens were fixed with 4% PFA, embedded in paraffin, sectioned at 5 µm, and stained with HE, safranin O, and aggrecan. In N-TEC group, histological analysis of graft sections showed morphology approaching that of native cartilage, which was confirmed by quantitative T1 measurements showing two dinstinct layers in post-contrast images. In N- CAM group, weaker safranin O and aggrecan staining were observed, which was reflected in the MRI images where no distinct layers were noticeable.Gadolinium-enhanced MRI relaxometry has the potential to be used for the characterization of cartilage tissue grafts as an alternative to standard methods.
