In this work we examined the incorporation of the peptidoglycans and adamantyltripeptides into the liposomes with the aim to possibly increase and optimize their therapeutic efficacy. Our studies concern two groups of immunomodulators: a) peptidoglycan monomer (PGM, b-D-GlcNAc-(1®4)-MurNAc-L-Ala-D-isoGln-meso-A2pm(eNH2)-D-Ala-D-Ala) and its two semisynthetic derivatives, Boc-L-tyrosyl-peptidoglycan monomer and (adamant-1-yl)-acetyl peptidoglycan monomer; b) synthetic adamantyltripeptides comprising a part of peptidoglycan structure (D- and L-(adamant-2-yl)-Gly-L-Ala-D-isoGln). The large multilamellar negatively charged vesicles using egg lecithin were prepared. The liposome preparations were chromatographed on the Sephadex G-50 or centrifuged in ultracentrifuge. The entrapment of peptidoglycans and the stability of respective preparations were followed in two ways: by the use of 14C-labelled peptidoglycans or by HPLC. The influence of the chemical structure (lipophilic-hydrophilic) on the amount of entrapped material into liposomes will be shown.
