Long-nosed viper (V. ammodytes) is one of the most poisonous snakes, that is widely distributed through central Europe. Specific antiserum, that is produced by gradual immunisation of big animals (horse, donkey, sheep), is the only available therapeutic drug for curing snake venom poisoning. The aim of our work was to establish a mice model for studying immune reaction (IR) specific for the snake venom, that is a toxin and an immunogen at the same time, and to monitor the influence of experimental adjuvants, peptidoglycan monomer GlcNAc-MurNAc-L-Ala-D-isoGln-mesoDAP(wNH2)-D-Ala-D-Ala (PGM) and L-(adamant-2-il)-Gly-L-Ala-D-isoGln (AdTP2), on this reaction. Optimal immunisation protocol was found by varying the amount of the venom dose and the schedule of immunisation. The amount of the antigen was scaled down starting with the maximal quantity of the venom that is not subcutaneously lethal for mice. The immunisation schedule, we found to be optimal, included four s.c. immunisations in three week intervals. The specific IR was monitored by ELISA measuring venom-specific IgGs (qualitatively and quantitatively). The selected sera with high titres of specific antibodies were tested for their ability to neutralise snake venom. The stimulatory effect of adjuvants, PGM and AdTP2, was demonstrated using suboptimal doses of venom.
