Sažetak
Fascioloides magna or large American liver fluke is a trematode parasite that can be found in parenchyma or in pseudocysts in livers of domestic and wild ruminants. Here, we present a comprehensive GC-MS based method of identifying and quantifying metabolites of this parasite and red deer (definitive host) tissues important for parasite feeding. A total of 40 metabolites were quantified using GC-MS. Data confirmed significant differences in metabolism between adult and juvenile flukes. Adult flukes had increased concentrations of l-valine, l-proline, l-serine, l-aspartic acid, l-histidine, palmitoleic acid, linoleic acid, cholesterol and phosphoric acid and decreased concentrations of l-glycine, glycerol and glucose. Multivariate Principal Component Analyses (PCA) revealed capsule fluid as the richest source of almost all amino acids in comparison to other tissues, while the liver tissue was more related to glucose and glycerol content and adult flukes to cholesterol content. PCA analyses of lipid components revealed four clusters: liver tissue with high content of polyunsaturated fatty acids, capsule fluid as the richest source of linoleic and linolenic acid, blood as the richest source of oleic acid and cultured, adult and juvenile flukes with increased de novo lipogenesis. Insight into metabolome of Fascioloides magna is an interesting approach to better understand the host-parasite interaction and may provide opportunities for identification of markers essential for the diagnosis of diseases and for the monitoring of new therapeutics.
