Sažetak
Congenital human cytomegalovirus (HCMV) infection is the most common viral cause of long-term neurodevelopmental sequelae, including mental retardation, microcephaly and sensorineural hearing loss. Central nervous system (CNS) damage is the major cause of these long term sequelae. The pathogenesis of CNS disease in congenitally infected human infants remains poorly defined. As HCMV does not cross the species barrier, we employed a mouse model in which newborn mice are infected by intraperitoneal inoculation of mouse cytomegalovirus (MCMV). We have demonstrated that MCMV model recapitulates all key characteristics of the human disease, including hearing loss. Following infection, the virus disseminates to the CNS and replicates in the brain parenchyma. CNS infection leads to inflammation, activation of resident microglial cells and the recruitment of peripheral immune cells. The role of innate and acquired immune cells in the pathogenesis of MCMV infection in the brain of newborn infected mice will be the major subject of my talk.
