Sažetak
Peroxisome proliferator activated receptor- (PPARG) and adiponectin (ADIPOQ) play important role in lipid and glucose homeostasis, insulin resistance and adipogenesis, and their gene variants could be important markers for understanding the genetic basis of metabolic syndrome (MetS). The aim of the study was to estimate possible associations of PPAR Pro12Ala and ADIPOQ -11377C>G and -11391G>A polymorphisms with MetS and its traits. Since many gene are involved in the regulation of MetS phenotypes or phenotypes of some of its traits we also tested interplay between PPAR and ADIPOQ variants. Study included 149 subjects, aged 20-35 years. Biochemical and anthropometric parameters were measured by standard methods. The ADIPOQ gene promoter SNPs were genotyped by real-time PCR. Genotyping of PPAR Pro12Ala was performed using PCR-RFLP method. In the total sample ADIPOQ promoter SNPs were associated with waist circumference, where ADIPOQ -11377GG and ADIPOQ -11391GA increased risk for the development of abdominal obesity (OR 5.57 and OR 3.37, respectively). ADIPOQ -11377G and -11391A allele and haplotype were associated with increased TG. Furthermore, the G allel at position -11377 as well as the A allele at position -11391 increased the risk for MetS (OR 2.00 and OR 2.43, respectively). Test of overall associations showed that specific haplotype -11377G-11391A had the strongest association with development of MetS (OR 4.09). Significant interaction was detected between PPAR Pro12Ala and ADIPOQ -11377C>G that affected glucose concentration (p=0.043) in the manner that Pro/-11377G contributed to the highest values of glucose (specific p=0.036). Interaction between PPAR Pro12Ala and ADIPOQ -11391G>A also affected glucose concentration (p=0.013) in the manner that Ala/-11391A contributed to the lowest values of glucose concentrations (specific p=0.008). Association was detected between PPAR Pro12Ala and ADIPOQ -11391G>A genotypes and LDL-C levels (p=0.032). In conclusion, PPAR gene variants can, in interaction with ADIPOQ as target gene, modulate physiological processes leading to the development of MetS, and they may be useful biomarkers for estimation of MetS risk in a young population.
