Sažetak
Diabetes mellitus (DM) is a metabolic disorder of the endocrine system with a rising of incidence in the next twenty years. Both main forms of diabetes (type 1 and 2) can lead to micro- and macrovascular complications including ischaemic heart disease, stroke, peripheral vascular disease and other diseases. Type 2 diabetes (T2D) is the most frequent type (>85%) of total diabetes prevalence. Growing evidence based on animal studies supports the anti-diabetic effect of some dietary polyphenols (DP) as one of therapy proposed for diabetes management and prevention of its long-term complications. The hypoglycemic effects of DPs may be related to inhibition of carbohydrate digestion and glucose absorption. Polyphenols may suppress glucose release from the liver, and ameliorate glucose uptake in peripheral tissues by modulating intracellular signalling and also can inhibit advanced glycation endproduct (AGE) formation. Although many epidemiological studies support favourable effects of polyphenol-rich diets in preventing and managing T2D, little is known about polyphenols in the co-treatment of T1D. The objective of this study was to determine the effect of olive leaf extract (OLE) rich in oleuropein on possible T1D treatment in diabetic rats. T1D was induced by a single i.p. administration of streptozotocin (SZT, 65 mg/kg of BW) in male Wistar rats divided into five groups: control, SZT-treated and three dose-dependent OLE post- treated groups. The blood glucose was monitored at set time intervals. Twenty-four hours after the last dose of OLE or saline, rats were sacrificed and liver and kidney were removed. Protein concentrations and alpha-glucosidase activities were determined in liver and kidney homogenates. Data were expressed as the mean ±SD. Differences between groups were assessed by a nonparametric Kruskal–Wallis test. Differences with P under 0.05 were considered to be statistically significant. The results show that OLE was dose-depending alpha-glucosidase inhibitor both in rat kidney and liver, therefore, can be effective in co- treatment of T1D and prevention of long-term complication induced by DM.
