Sažetak
Acute and chronic stress can induce the production of reactive oxygen species that could lead to oxidative stress, which is recognized to be one of the key pathophysiological mechanisms in liver disease development. The aim of this study was to estimate the impact of acute and chronic stress on the oxidative stress development and antioxidative status of the rat males liver as well as non-ovariectomized (NON-OVX) and ovariectomized (OVX) rat females liver. Sixty 16-week-old Sprague-Dawley rats were included in the study and divided into eight groups: males, NON-OVX and OVX females that were exposed to acute stress ; males, NON-OVX, and OVX females that were exposed to chronic stress ; and males and NON-OVX females as a control groups. As an indicator of liver oxidative damage, lipid peroxidation levels expressed in terms of thiobarbituric acid reactive substances (TBARS) were determined, while liver antioxidative status was determined by catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GPX), glutathione S-transferase (GST) activities and reduced glutathione (GSH) content. Both acute and chronic stress significantly increased TBARS content in liver of males, NON-OVX and OVX females relative to control groups. Results showed that acute and chronic stress lowered antioxidant defense capacity, as evaluated by the significant decrease in the GSH content and decrease of some antioxidative enzyme activities. Both acute and chronic stress decreased CAT activity in all tested groups when compared to control groups. SOD activity was lower in all acutely stressed groups relative to controls, while chronic stress resulted in lower SOD of OVX females. Furthermore, GPX and GST activities were decreased in NON-OVX under acute stress. Under chronic stress GPX activity was decreased in OVX and NON-OVX females, and GST activity was decreased in males, NON-OVX and OVX females. The analyzed data indicate that acute and chronic stress induced oxidative damage in liver and decreased liver antioxidant defense capacity. The oxidative and antioxidative responses to acute and chronic stress were sex specific, while the extent of the oxidative stress was the highest in the OVX females.
