Sažetak
The current issues in radiotherapy are dealing with minimizing normal tissue toxicity and at the same time keeping the levels of radiotherapy effectiveness. Radiomodulation is one of the ways how clinicians can achieve such effects. The use of radioprotectors in radiotherapy is today limited to only one approved agent-amifostine whereas there are many agents that are used as radiomitigators. Chlorophyllin (CHL) is a derivate of natural plant pigment chlorophyll and it is used in food production as a food dye. It has also shown several beneficial effects on different experimental models which makes it interesting molecule to study its radiomodulating properties. Using human peripheral blood lymphocytes as a model system we examined the possible radioprotective properties of CHL. From the irradiated samples pre-treated for 2 h with CHL concentrations of 10 and 100 μg/mL, using comet assay, we managed to detect 19.31 % and 27.36 % reduction in DNA damage, respectively, compared to 5 Gy irradiated sample. However, only the latter concentration managed to reach statistical significance. At the same time CHL alone, at tested concentrations, did not induce DNA damage. Taken together, obtained results indicate that CHL is a molecule with potential radioprotective properties which at the same time is not toxic to human cells in vitro. Therefore, further studies are required in order to detect optimal radioprotective CHL concentration, its mechanisms of action and comparison to known radioprotectors.
