Sažetak
The oocyte is surrounded by follicular fluid originating mainly from granulosa and theca cells and from blood. Follicular fluid contains components important for normal oocyte maturation and ovulation. For these processes oxidative-antioxidative balance is required. The aim of this study is to determine oxidative stability and antioxidant potential of bovine follicular fluid influenced by breed and age. The research was conducted on two groups: Holstein Frisian (HF ; heifers N=7, aged 1.5±0.3 yr ; cows N=19, aged 4.8±2.5 yr) and Simmental or crossbreed with Simmental (ScS ; heifers N=12, aged 1.4±0.3 yr ; cows N=11, aged 7.2±2.4 yr). Follicular fluid samples were obtained from three different sized follicles (<5 mm, >5 mm and >10 mm). Reactive oxygen metabolites (ROM) and biological antioxidant potential (BAP) concentrations in follicular fluid were determined. Follicular fluid BAP concentrations in ScS heifers and cows were significantly lower in follicles >10 mm compared to follicles <5 mm and >5 mm. In follicular fluid of HF cows, BAP and ROM concentrations were significantly lower in follicles >10 mm compared to that in smaller follicles. Follicular fluid BAP concentration showed breed-dependent significant differences in follicles >5 mm. Significant age-dependent differences in BAP concentration in all follicles of ScS breed were noticed. In HF animals, significant age-dependent changes were found in BAP concentration in follicles >10 mm and in ROM concentrations in follicles <5 mm and >5 mm. The results indicate oxidative stability of bovine follicular fluid despite breed and age differences. The changes observed in follicular oxidative stability and antioxidative capacity were associated with oocyte development and appeared to be applicable as biomarker of oocyte fertilization ability.
