Sažetak
The aim of study was to investigate whether low level pesticides concentrations affect cell vitality under laboratory conditions. Selected concentrations of four different pesticides are considered equivalents to those corresponding to human population real-life exposures. Two out of four were insecticides chlorpyrifos and imidacloprid and the others were herbicides terbuthylazine and glyphosate. Concentrations of insecticide chlorpyrifos were calculated to be 3, 0.026, 0.000623, 0.016 μg/ml. Calculated concentrations for imidacloprid were 3, 0.13, 0.3146, 0.1 μg/ml. Herbicides terbuthylazine was tested at concentrations of 3, 0.008, 0.0008, 0.000583 μg/ml and glyphosate at 3, 2.91, 0.5 μg/ml. Continuous cell culture of human liver hepatocellular carcinoma cells, HepG2 was used. Six cell samples per concentration were exposed to selected pesticides throughout 4 hours. Negative- and positive control cell samples were included into the study. Pesticides concentrations were estimated to correspond to the occupational (OEL) and residential (REL) exposure level, or acceptable daily intake (ADI). After pesticides treatment cellular dehydrogenases activity was determined by colorimetric assay (WST-8). In comparison to the matched control cell samples, the viability of treated cells remained within physiological control values regardless of applied concentrations of pesticides solutions. In conclusion: selected concentrations of chlorpyrifos, imidacloprid, terbuthylazine and glyphosate did not affect cellular dehydrogenases activity. Low level pesticide concentration did not provoke in vitro cytotoxic effect. This work has been supported by Croatian Science Foundation under the project 8366.
