Sažetak
Objectives. Bovine mastitis is the main cause of economic losses in dairy industry and food safety concerns due to antimicrobial use. Although there are many treatment options avaliable, there is a lack of efficacious solutions concerning the subclinical mastitis. Many studies of treatment options proposed some alternatives to conventional bovine mastitis treatment solutions. Propolis is a resionous mixture that honey bees collect from tree various botanical sources, interesting due to its numerous biomedical effects ; immunomodulating, antioxidative and antimicrobial, among others. The main goal of our work is the research and development of the innovative concept of intramammary nonalcoholic formulation of propolis as an alternative to existing prevention and treatment of subclinical mastitis (with antibiotics). The development is conducted parallel with the research in dairy cows and goats together with veterinarians and livestock owners. Materials and methods. Main milestones of our research are: analysis of the formulation polyphenol composition (using GC-MS method), in vitro study of its antimicrobial efficacy against common mastitis pathogens (using agar dilution method on Mueller-Hinton agar), as well as clinical trials on Holstein cows and Saanen goats during dry period and lactation. Health and productivity, the incidence of subclinical and clinical mastitis as well as milk quality, oxidative stress and various metabolic parameters are monitored through clinical observation of animals. Results. Preliminary results show good in vitro activity of nonalcoholic propolis formulation against selected gram-positive bacteria (Staphylococcus aureus, coagulase-negative staphylococci), with minimum inhibitory concentrations (MICs) of propolis in the range from 16 to 64 mg/L. Significantly lower activity was determined for E. coli and Pseudomonas aeruginosa with MICs higher than 5000 mg/L. Clinical safety trials revealed different tissue reaction in cow vs. goat udder towards propolis formulation, which lead to the modifications of the original composition that will be tested in further trials.
