Sažetak
Background and Purpose: Since ordinary brown naphtalane (OBN) contains aromatics including those which are considered carcinogens, a non- aromatic version of naphtalane (NAN) was invented. Its composition was found favorable and it was admitted for a clinical study which proved NAN to be effective in the treatment of psoriasis vulgaris and not to be hemato-, nephro- or hepatotoxic. Thus, it was estimated to be a promising potential natural medium deserving a thorough research, including its chemical composition. The aim of the paper is to improve the understanding of the NAN composition. Potentially bioactive components were studied in great details. Material and Methods: The sample of NAN was a non-colored transparent unctuous liquid. It was declared not to be toxic or irritant. In order to prove its non-aromatic character the DAB 10 procedure was applied with the use of UV/VIS In this study GC-MS served in the focused study of steranes. Mass fragmentograms m/z 372, m/z 386 and m/z 400 were used to study cholestanes, ergostanes and stigmastanes according to their molecular ions. GC was applied to examine the mixture complexity of NAN. Conclusion: UV/VIS spectra displayed a clear proof of non-aromatic character of NAN. According to the GC chromatogram and its non-aromatic character, NAN is the rather complex mixture of a large number of saturated hydrocarbons. The focused mass fragmentograms permitted a sharp discrimination among cholestanes, ergostanes and stigmastanes. Also, four of the isomers in each of the homologs were clearly discriminated. Conformation 14(H)17(H)20R was found to be the most intense one in each studied sterane. Ergostane and stigmastane skeleton might be compared to the skeleton of provitamins D. There are additional natural and synthesized bioactive compounds with similar skeletons. The obtained results cannot solve the question what makes NAN bioactive, but shed some light on this complex subject.
