Sažetak
Aim: High salt diet (HS) is a well known risk factor for cardiovascular diseases characterized by sustained inflammation. Several studies reported that HS can promote development of autoimmune diseases in the presence of Th17-inducing factors, suggesting its role in the T-cell activation and differentiation. In this study we have examined the effects of short term HS on the rate of activated CD4 and CD8 T cells, and Treg in the peripheral blood and spleen of naive rats. An additional aim was to test if the observed effects are related to increased ROS production found during HS. Methods: 12-weeks old male Sprague-Dawley rats (N=5/group) were fed with standard rat chow or high salt diet (0.4% or 4% NaCl, respectively) during 7 days. One group of HS rats received a SOD mimetic (TEMPOL) in drinking water ad libidum. Spleen and peripheral blood mononuclear cells (PBMC) cells were stained with antibodies against CD4, CD8, CD25 and Foxp3 and analysed by flow cytometer. Results: Rats fed HS had significantly reduced frequencies of activated CD4+CD25+ helper and CD8+CD25+ cytotoxic T cells, as well as Treg among PBMC, compared to control group. TEMPOL almost completely reversed the rate of these cells to control values. Conclusions: Results of this study suggest that HS can affect T cell activation and Treg expansion/differentiation. In addition, these effects might be related to increased ROS production during HS.
