Sažetak
Aim: Hyperbaric oxygenation (HBO2) could modulate the mechanisms of endothelium dependent vasorelaxation of conductive vessels in healthy and diabetic animal. The aim of this study was to evaluate the effects of HBO2 on gene expression of cyclooxygenase 1 and 2, NOS and CYP P450 isoforms: CYP4A1, CYP4A3 and CYP2J3. Methods: 12 weeks-old male rats were used- control, diabetic (DM), and control-HBO2 and DM-HBO2 group of rats that underwent HBO2 treatment (120 minutes in duration, at 2.0 atm for 4 consecutive days, 5th day sacrificed), N=5. Diabetes was induced by streptozocin (in 6 weeks old animals). Aortas were collected for mRNA expression analysis of COX 1 and 2, NOS and CYP P450 genes. Data were normalized with HPRT1 gene expression. Results: COX1 and eNOS genes were up-regulated in DM+HBO2 compared to DM and control groups. Relative expression of CYP4A1 gene was increased in DM+HBO2 group compared to control+HBO2 group and CYP4A3 gene was down- regulated in control+HBO2 and DM groups compared to control group. Conclusions: Our study shows that HBO2 treatment in diabetes changes expression of COX1 gene. Increased CYP2J3 and CYP4A1 expression in DM rats and down-regulation of their expression in control rats by HBO2 treatment may explain our previous results on beneficially effect of HBO2 to vascular response in disease condition.
