Sažetak
The myelodysplastic syndrome (MDS) comprises a heterogeneous group of clonal bone marrow disorders characterized by abnormal differentiation and maturation of myeloid cells, bone marrow failure and a genetic instability with enhaced risk to transform to secondary leukemia. As ACE has been shown to enhance regrutation of bone marrow stem cells into the S-phase of the cell-cycle, we choose to investigate its role in dysfunctional haemopoiesis of the MDS. After establishing of methods for DNA isolation from bone marrow smears, amplification of specific DNA region and immunohystochemical staining of ACE protein in bone marrow biopsies, ACE I/D genotype distribution, ACE expression on bone marrow cells and their correlation with biochemical parameters of MDS patients was done. No statistically significant difference in genotype distribution between MDS patients and controls has been found. However, significantly higher ACE expression as well as the number of cells which express it in the bone marrow of MDS patients have been observed. By multiparametric analysis of biochemical parameters with ACE expression and ACE genotype statistically significant negative correlation between ACE expression and number of platelets has been found. The ACE gene polymorphism had no significant influence on MDS patients survival.
