Sažetak
Population of Black Slavonian pig, once highly valued and almost the only breed in Slavonia, has significantly decreased over the years. With modest demands of nutrition and housing, they are intended for the breeding in the open. Research and preservation of this unique genetic potential are priority because of its quality. It is characterized by high quality meat, meant for processing in traditional dried meat products. This breed belongs in the middle fertile breed with 6-8 piglets per litter. But the fertility of sows, as one of the most important economic characteristics, is quite uneven, and thus represents a very important trait for the selection. Some researchers have established that the polymorphism of prolactin receptor gene (PRLR-gene) can be correlated with fertility of sows and thus used as a potential genetic marker in selection. The aim of the study was to isolate DNA from hair follicle samples of 15 Black Slavonian sows and genotype polymorphism of PRLR receptor. After DNA isolation with commercial kit One-tube Hair DNA Extraction Kit (Bio Basic Inc., Markham Ontario, L3R 8T4 Canada) multiplication of PRLR sequence was performed using specific primers (SIGMA, Aldrich). Obtained PCR products of 163 bp were subjected to restriction enzyme AluI in order to determine the polymorphism of PRLR-gene. For each genotype were obtained following combinations of restriction sequence: AA - sequences of 85, 59 and 19 bp, BB - 104 and 59 bp, and AB - 104, 85, 59 and 19 bp. The results have shown even distribution of AA (40%) and BB (40%) homozygotes, while AB heterozygote was less frequent (20%).
