Sažetak
Phosphate is often the growth limiting factor in natural waters. Abiotic decomposition of organic phosphorous compounds is very slow under environmental conditions. Organic phosphorus compounds in water, either of biological origin or as products of human activities, can not be directly assimilated by planktonic organisms. Inorganic phosphate becomes available only upon enzymatic hydrolysis outside the cell. This reaction represents the key stage in phosphorus cycling in natural waters. It has been established that a significant number of organophosphorus compounds can be hydrolyzed by alkaline phosphatase (E.C. 3.1.3.1. ALP), an enzyme present at the outer side of the cell membrane. Natural substrates for ALP are unknown, as well as their affinity or rate of hydrolysis in vivo. The influence of the organic and inorganic phosphate concentration on species selection, and the mechanism of phosphate transport into the cell remain unresolved questions. The aim of this study was to improve the method for the determination of ALP activity in seawater samples. Since the synthesis of ALP depends on the phosphate concentration, the measured activity has been used as an indicator of phosphorus deficiency in a phytoplankton community. For that purpose the concentration of inorganic phosphate has been determined, simultaneously with the phytoplankton analysis of the same seawater samples.
