Sažetak
Trefoil factor family 3 (TFF3) has been suggested as a good urinary biomarker for early detection of kidney tubular injury. We measured TFF3 expression in an animal model of chronic kidney disease (CKD) caused by unilateral nephrectomy (UN). 36 female Sprague Dawley rats were randomized into the three groups: unilaterally nephrectomized (A), sham operated (B) and naive rats (C). 12 months after UN the rats were sacrificed and the kidneys were taken for immunohistochemical analysis. Kidney function was followed up by repeated measurements of serum creatinine, sodium and potassium concentrations. Impairment of kidney function and development of CKD was proven by significantly increased serum creatinine concentrations in UN rats when compared to both control groups (A=40.3±5.9 ; B=29.1±4.4 ; C=29.6±2.9 ; p≤0.001. ANOVA). In the remaining kidneys of UN rats, the HE and PAS stained sections analysis revealed kidney tissue damage, including the loss of functional tissue, fibrosis, and lymphocyte infiltration. TFF3 was detected in all parts of the kidney tubular segment. However, its expression did not differ between remaining kidneys in UN rats and both control groups, nor to the young kidneys removed during the nephrectomy. Thus, our results suggest that the kidney expression of TFF3 is a weak biomarker of CKD.
