Sažetak
The genus Mycobacterium (M.) comprises bacteria that are heterogeneous in terms of metabolism, growth, environmental niche, epidemiology, pathogenicity, geographic distribution and disease association. Most of these nontuberculous mycobacteria (NTM) are environmental organisms typically acting as opportunistic human pathogens. During 2013, bacteriological investigation was carried out on presence of different NTM in cattle and wild boars in Croatia. Wild boars were used as species indicator of ecosystem and cattle positive on tuberculin skin test were used as species involved in food production. Bacteriological isolates were identified using routine molecular methods: conventional PCR, GenoType Mycobacterium CM assay (Hain, Germany) and GenoType Mycobacterium AS assay (Hain, Germany) and biochemical testing. In addition six wild boar isolates were shipped to NRC for Mycobacteria, Research Center Borstel in Germany where they were analyzed by sequencing of multiple genes, including 16S rRNA and rpoB and by 16S-23S rRNA internal transcribed spacer (ITC) sequencing. Using molecular methods seven cattle isolates were identified as M. avium (1 isolate), M. celatum (1 isolate) and Mycobacterium sp. (5 isolates). Five isolates identified as Mycobacterium sp. were furthermore analyzed by classical biochemical testing and were identified as M. flavescens, M. chitae, M. vaccae and Mycobacterium sp. Seventy wild boar isolates were identified using molecular methods. Isolates were identified as M. fortuitum (4 isolates), M. intermedium (1 isolate), M. celatum (5 isolates), M. avium (1 isolate) and Mycobacterium sp. (59 isolates). A total of 41 isolates identified as Mycobacterium sp. were furthermore subjected to classical biochemical testing by which two more species were identified as M. vaccae and M. agri. For 39 isolates biochemical testing is still in progress. Multigene sequence-based typing of 6 wild boar isolates was successful for 5 (83.3%) isolates and they were identified as M. vaccae, M. neoaurum, M. phocaicum, M. arupense (2 isolates) and typing of one isolate is still ongoing. Performed even on limited number of isolates, multigene sequence-based typing was more successful in identification of NTM than molecular tests used for routine analysis in our laboratories. Identification of NTM presence in different niches is important epidemiological information because of rising virulence and resistance, particularly in some species.
