Sažetak
BMP are expressed in bone marrow stromal cells and involved in regulation oh hematopoiesis. The aim of this investigation was to explore expression of bone morphogenetic proteins, their receptors and BMP second messengers in long- term culture of human bone marrow stromal cells. Hematopoietic activity was present throughout the investigation period. RNA was isolated from cultivated cells using a TRIzol reagent ; cDNA was then synthesized and RT-PCR performed using primers for BMP-2 to 7, ALK-2, 3 and 6 and Smad-1, 4 and 5. The results indicate that bone marrow stromal cells produce mRNA for ALK-2 and 6, Smad-1, 4 and 5, and both mRNA and protein BMP-3, 4 and 7. Since hematopoietic cells have BMP receptors, a paracrine mechanism in regulating hematopoiesis is suggested.
