Sažetak
LeuRS efficiently activates and transfers norvaline to tRNALeu and thus relies exclusively on post-transfer editing to eliminate norvaline from the genetic code. In sharp contrast, high level of discrimination against isoleucine is achieved in the synthetic reaction, thereby obviating the need for post- transfer editing. CP1 domain is capable of rapid hydrolysis of both Ile- and Nva-tRNALeu. In spite of slow transfer of isoleucyl moiety to tRNALeu, LeuRS does not display tRNA dependent pre- transfer editing. The main function of E. coli LeuRS CP1-editing domain is to exclude norvaline from the cellular proteome.
