Sažetak
Introduction: The objective of this study was to investigate the effect or sodium lauryl sulphate (SLS) on the levels of natural moisturizing factors (NMFs) in the stratum corneum (SC). SLS is a common ingredient of soaps and cosmetic products and is frequently used as a model compound in experimentally induced skin irritation. NMFs are present within the corneocytes and play a key role in the maintenance of epidermal hydration. They are mainly formed from filaggrin proteolysis to small, hygroscopic molecules including amino acids. Reduced levels of NMF lead to dry skin. Material and methods: On three consecutive days 20 volunteers were dermally exposed for 4 hours to water, 0.1%, 0.25% and 0.5% SLS on one volar forearn using Finn patches of 18 mm Ø . Two assess the irritation response the following parameters have been measured before and after exposure to water and SLS on day 2, 3 and 4. 1.Transepidermal water loss (TEWL), pH and erythema 2.Levels of filaggrin degradation products (including 2-pyrolidone-5-carboxylic acid, histidine and urocanic acid) in the stratum corneum harvested by using adhesive tape DSquame). NMF levels in the stratum corneum have been determined by using an HPLC method. Results: Dermal application of SLS resulted in concentration and time dependent irritant reaction, reflected significant increase in TEWL, erythema and skin pH. Dermal exposure to SLS was associated with reduced levels of the NMFs in the SC ; the effect on the NMFs was correlated with skin barrier damage as assessed by TEWL. Interestingly also exposure to water resulted in gradual decrease of the NMFs. Conclusion: Dermal exposure to water and SLS led to reduced levels of the NMF components in the uppermost layer of the skin, which can lead to dry, cracky skin and in the case of chronic exposure contribute to the development of irritant contact dermatitis.
