Sažetak
The wine yeast Dekkera bruxellensis is considered to be the main cause of wine spoilage world wide. This yeast generates off-flavours during the maturation process, thereby causing immensive financial losses. D. bruxellensis shares several common features with Saccharomyces cerevisiae, like production of alcohol under aerobic conditions, ability to grow without oxygen and a petite-positive character, but these two yeasts are not closely related. D. bruxellensis has very dynamic chromosomes. We attempted to study D. bruxellensis replication elements. For this purpose genomic libraries based on two strains were prepared and a transformation protocol developed. Surprisingly, from two different strains only three different loci, named CIGO1, CIGO2 and CIGO3, could support autonomous replication of the introduced plasmids. When we examined the recently sequenced genome sequence we could find that two CIGO loci are over-represented in the genome and thereby present in more than one copy per genome. We speculate that the CIGO elements may be involved in the re-shuffling of the chromosome.
