In this study hydroxylamines have been examined as oxygen-sensitive procontrast agents for in vivo MRI. Hydroxylamines obtained by the reduction of stable nitroxides were injected subcutaneously in one hind limb of rat, while in the other hind limb a physiological saline was injected as a control. Images of the transversal sections of rat were recorded and contrast enhancement due to oxygenation of injected hydroxylamines in hind limb was shown. This can be explained by the chemical transformation of the nonparamagnetic hydroxylamine (procontrast agent) to the paramagnetic nitroxide (contrast agent), which shortens proton relaxation times. No oxidation of hydroxylamines was observed if the limb was kept hypoxic. These observations demonstrate the feasibility of a method of selectively observing oxygenated versus hypoxic regions in MR imaging.
