Sažetak
Vitamin C ili askorbinska kiselina jedan je od osnovnih nutrijenata u ljudskoj prehrani. To je ketolakton, sličan glukozi, strukturne formule 2, 3-endiol-L-gulonska kiselina-γ-lakton. Metodologija analize askorbinske kiseline napredovala je od biološkog istraživanja do instrumentalno naprednih spektrofotometrijskih, fluorometrijskih, elektrokemijskih i kemiluminiscentnih metoda. Tekućinska kromatografija s UV-VIS, fluorescentnim ili elektrokemijskim detektorima pokazala se kao vrlo efikasna metoda za odjeljivanje različitih izomera askorbinske kiseline. Cilj ovoga rada bio je usporediti dvije metode analize, HPLC i UV/VIS, primijenjenih pri analizi askorbinske kiseline u šumećim tabletama vitamina C dvaju proizvođača Naturel i Encian. UV/VIS analiza askorbinske kiseline provedena je pri valnoj duljini od 247 nm, a odjeljivanje na HPLC uređaju provedeno je izokratnom elucijom s mješavinom otapala metanola i vode (95:5, V/V). Detekcija askorbinske kiseline provedena je na UV/VIS detektoru. Kalibracijski pravci kod obaju metoda HPLC i UV/VIS pokazuju vrlo visoke koeficijente korelacije (R2) više od 0, 99 ukazujući na proporcionalno povećanje koncentracije askorbinske kiseline u linearnom području mjerenih koncentracija u ovisnosti o apsorbanciji kod UV/VIS metode odnosno površine ispod pika kod HPLC metode. Rezultati analize askorbinske kiseline UV/VIS metodom pokazuju prosječno 6, 7% odstupanje od mase deklarirene na proizvodu, dok su kod HPLC anlize ta odstupanja nešto viša i iznose 15, 7%. Dobro slaganje rezultata UV/VIS analize s količinama deklariranim na proizvodima ukazuju da je ova metoda brza, jedostavna, precizna i točna te primjenjiva za kvantitavno određivanje askorbinske kiseline u pripravcima vitamina C. Viša odstupanja rezultata analize askorbinske kiseline dobivenih HPLC metodom u odnosu na UV/VIS metodu ukazuju da je ovu metodu potrebno još dorađivati. Usprkos tome HPLC metoda pokazala se pogodnom za analizu askorbinske kiseline u šumećim tabletama zbog svoje brzine, osjetljivosti te ponovljivosti u rezultatima.

Vitamin C or ascorbic acid is one of the main nutrient in the human diet. It is ketolactone, similar to glucose, with the structural formula 2, 3- endiol-L-gulonic acid-γ-lactone. Methodology of the ascorbic acid analysis has advanced from the bioassay to the instrumentally advanced spectrophotometric, fluorometric, electrochemical, and chemiluminescence methods. Liquid chromatography with UV-VIS, fluorescent or electrochemical detectors proved to be an excellent tool for separating the different isomers of ascorbic acid. The aim of this study was to compare two methods of analysis, HPLC and UV/VIS, by analyzing ascorbic acid in effervescent tablets of vitamin C produced by two manufacturers Naturel and Encian. UV-VIS analysis was carried out at a wavelength of 247 nm, while HPLC separation was conducted by isocratic elution with a solvent mixture of methanol and water (95:5, v/v). Detection of ascorbic acid was performed with UV/VIS detector. Calibration curves in both HPLC and UV-VIS analysis showed a high correlation coefficients (R2) higher than 0.99, indicating a proportional increase of the ascorbic acid concentration in the linear area of the measured concentration with absorbance in UV -VIS method or area under the peaks in HPLC method. Results of the ascorbic acid analysis obtained by UV/VIS method showed deviation from the mass declared on the tablet product of 6.7% deviation (in average) while deviation achieved by the HPLC analysis was higher (15.7%). The deviation in results obtained by UV/VIS analysis indicates that this method is fast, simple, precise and accurate as well as applicable to quantify the mass of the ascorbic acid in the vitamin C products. Higher deviation of the results obtained by HPLC in comparison to UV/VIS method showed that this method needs improvement. Nevertheless HPLC method proved to be suitable for the analysis of ascorbic acid in the effervescent tablet due to short time of analysis, sensitivity, and results reproducibility.
