Stemness genes Oct and Nanog were recently suggested by our and other studies to be involved in development of germ cell tumours (GCT). Germ cell like tumours can be experimentally obtained by cultivating gastrulating mouse embryos in vitro. EsiRNAs are constructs able to partake RNA interference pathway of the cell to induce mRNA degradation i.e. inhibit the expression of various genes. The aim of this study was to determine whether tumour growth can be inhibited by RNAi suppression of stemness genes Oct and Nanog. Therefore, esiRNAs targeting coding regions of mouse Nanog, mouse Oct4 or GFP were synthesized. 7, 5–days-old C3H mouse embryos were microsurgically isolated and cultivated at the air-liquid interface for 7 days where they developed to a teratoma-like structure. Specific esiRNAs in transfecting agent Lipofectamine 2000 were added to the medium. Embryos/GCTs were measured at the beginning of cultivation and for consequent 7 days. At the day 7, RNA was isolated and expression of Oct3/4 and Nanog was analysed by qPCR. EsiGFP treated served as a negative control since no esiGFP target exists in a mouse cell. Indeed, esiGFP group showed no statistically significant difference in growth when compared to controls. GCTs developed from esiOct and especially esiNanog treated embryos showed significant reduction in growth when compared to controls. EsiRNA induced drastic inhibition of stemness gene persistent throughout observed period. By this experiment we confirmed crucial role of stemness genes Oct and Nanog in GCT development and the utility of intervening in tumour growth by esiRNA treatment.
