Sažetak
In spite of our knowledge that human spermatozoa possess Cu, Zn superoxide dismutase (SOD), little known about the change in the levels of this enzyme asociated with sperm quantly and/or quality. SOD-like activity was determined in spermatozoa of oligozoospermic men characterized by reduced sperm concentation (less than 20x106 spermatozoa/ml) and in asthenozoospermic men exibiting reduced motility (less than 40 percent motile sperms). Normozoospermic samples with less then 20x106 spermatozoa/ml and more than 40 percent motile sperms served as controls. In some experiments Percoll gradient fractionation was used to separate the spermatozoa with high motility and and competent function (80 percent fraction) from those with diminished motility and poorfunctional capacity (40 percent fraction). SOD-like activity was measured by the degree of inhibition of the reaction between superoxide anion radicals generated by xanthine/xanthine oxidase system and I.N.T. tetrazolium salt, wherby a red formazan absorbing at 500 nm was formed. Significantly higher SOD-like activity was detected in 27 oligozoospermic samples compared with 58 normozoospermic samples (p 0.05). SOD-like activity in 12 asthenozoospermic samples from 40 percent gradient was significantly higher than those recorded in the same fraction of 18 normozoospermic samples (p 0.5). A positive relationship between SOD-like activity and experimentally-induced lipid peroxidation was detected in 54 infertile men (p 0.05). These findings suggest that the a higher level of SOD-like activity may reflect a defect in development or maturation of spermatozoa and, thereby,a decreased fertility potential.
