Sažetak
Fat free diets (FFD), which contain less than 1 % of total energy content as fats, are used as therapy in some lipid metabolism disorders and gastrointestinal diseases. Applied for a longer period of time, they can induce symptoms of essential fatty acids deficiency. We have started our studies in order to find out what kind of changes, if any, are caused in tissue fatty acid composition by feeding such fat free diet for a shorter period of time. Two groups of Wistar rats were fed for two weeks either standard rat chow as control diet (CD) or FFD prepared in our laboratory. Animals were sacrificed by heart vein puncture and total lipids were extracted by CHCl3:CH3OH mixtures from erythrocyte membranes and brain, heart and liver tissues. After methanolysis, the composition of fatty acid methyl esters was determined for all samples. The most pronounced alterations in fatty acid composition were found in liver tissue, while in other tissues modifications were of a lesser degree, but still evident. Generally, total lipids of rats on FFD contained less linoleic acid (C18:2, n-6), as well as other long chain polyunsaturated fatty acids (arachidonic, C20:4, n-6; eicosapentaenoic C20:5, n-3; docosahexaenoic, C22:6, n-3), when compared with animals on CD. At the same time, the content of monoenoic acids (palmitoleic, C16:1, n-6; oleic, C18:1, n-6) was increased, leading to the conclusion that endogenous synthesis of monounsaturated fatty acids was improved. Having in mind the importance of saturated vs. unsaturated fatty acids ratio for the structure and function of cell membranes, encouraging is the fact that essential fatty acids deficiency of such short term FFD can be compensated by endogenous synthesis.
