ω-3 Polyunsaturated fatty acids, besides having an essential role for the health of living organisms, undergo a large number of enzymatic transformations leading to compounds with potent anti-inflammatory and neuroprotective properties.1 Neuroprotectin D1 (NPD1) belongs to this family of lipid mediators and is found to be produced from docosahexaenoic acid (DHA) in retinal, fat and neural cells during oxidative stress induced diseases. NPD1 shows substantial neuroprotective, anti-inflammatory, regenerative and anti-apoptotic abilities.2, 3 Due to these properties NPD1 has become an interesting target molecule for the treatment of brain stroke and Alzheimer’s disease. Although two synthetic approaches were devised, 4, 5 new methods for very efficient, stereoselective and versatile syntheses of NPD1 and analogs are still needed. We present results of our efforts to design short and modular synthetic approaches to NPD1 and its analogs for various biological investigations.
