Sažetak
This study investigated two groups of GABAergic non-pyramidal cells: parvalbumin (PV) fast- skipping (FS) cells with morphology of basket or chandelier neurons and calbindin D28k (CB) positive cells marked as non fast-skipping (NFS) with vertically oriented axonal morphology in the cortex of wild type (WT), St3Gal2 (beta- galactoside alpha-2, 3-sialyltransferase 2) null, St3Gal3 (ST3 beta-galactoside alpha-2, 3- sialyltransferase 3) null and double null (DKO) mice. PV and CB neurons were quantified relative to all neurons (identified by NeuN) for: striatum (St), motor (M1), visual (V1), somatosensory (S1) and auditory (Au1) cortex. ST3Gal2 mice had a significantly lower proportion of CB positive neurons in all regions except Au1 cortex, whereas the proportion of PV positive neurons was lower in all sensory regions and St, but not in M1. ST3Gal3 mice had a significantly lower proportion of CB neurons in M1 and St, but the proportion of PV neurons was not decreased. Conspicuously, DKO mice had a higher density of NeuN positive cells in all regions except S1, and the proportion of CB positive neurons was equal to or higher than in wild type mice. We conclude that these sialyltransferase-null mice have a shift in the ratio of two subgroups of interneurons that are critical for cortical plasticity and have been implicated in the etiology of complex neuropsychiatric diseases like schizophrenia, autism, anxiety disorder and seizures. These findings are the first to report about CB and PV interneurons in these mice and may help to illuminate the role of sialic acid epitopes in the production and migration of interneurons.
