Extranodal marginal zone lymphoma of MALT (mucosa-associated lymphoid tissue) type is a low-grade B-cell lymphoma that originates from marginal zone cells of secondary follicles, which are generated in response to various types of chronic inflammations, i.e. infection of Helicobacter pylori. Epigenetic alterations associated with development of this type of lymphoma are not well studied at this point. So far, an increased DNA methylation level in specific genes was reported in relation to H. pylori infection. In addition, histone methylation marks were suggested to play an important role in development of MALT lymphoma. In this case study, we investigated the presence of the histone modification H3K27me3 (mark for repressive chromatin) in cells of gastric mucosa inflammation and tumor cells of gastric MALT lymphoma infiltrates. Sections from the gastric biopsies of 11 patients, performed at the time of diagnosis, as well as, after each cycle of therapy, were analyzed, and compared with normal tissue (intact mucosa tissue). Pearson’s χ2 test showed that cells with inflammatory changes, as well tumor infiltrates cells, had significantly different distribution of H3K27me3 (p=0, 017) compared to normal mucosa tissue. Samples exhibiting inflammatory changes had more H3K27me3 expression, while tumor cells expressed less H3K27me3 than expected. Our data indicate a trend of H3K27me3 decrease during the disease development. Since H3K27me3 is associated with transcriptional repression of many genes during normal development, stem cell maintenance and differentiation, our findings indicate possible reactivation of certain developmentally regulated genes as a basis for tumor development.
