Sažetak
Differentiation of plant cells has been described as a multistep process involving many morphological, histological and biochemical changes. Normal, hormone-dependent (N) and habituated, fully nonorganogenic (HNO) callus lines of sugar beet (Beta vulgaris L. altissima) have been intensively studied at the biochemical, morphological and physiological levels. They have been recommended as a model for studying plant cell differentiation. In addition to the N and H lines, we have established a tumour line (T) as a positive control for dedifferentiated, habituated callus. The expression of intra- and extracellular proteins in these three cell lines has been studied, and tissue-specific proteins and glycoproteins have been detected. In this study we compared the cellular structure and extracellular protein spectra of the N, HNO, and T cell lines in order to identify protein markers that might explain the distinct phenotypical characteristics of the cell lines. Therefore, the light and electron microscopic studies were undertaken, accompanied with electrophoretic separation and MALDI-TOF mass spectrometry analyses of extracellular proteins.
