Sažetak
OBJECTIVES: With the increasing evidence that decreased blood concentrations of n-3 polyunsaturated fatty acids (PUFA) have been associated with several neuropsychiatric disorders (i. e. depression, anxiety, panic disorder, schizophrenia), abundance of scientific papers focus on further investigation into lipid metabolism and prevalence of mental disorders. Based on recent studies suggesting dyslipidemia in individuals suffering from posttraumatic stress disorder (PTSD), an anxiety disorder, we examined the levels of PUFA and total cholesterol in red blood cells of combat veterans with a diagnosis of PTSD caused by war. METHODS: The study included 50 male war veterans diagnosed with PTSD according to DSM-IV and MKB-10 diagnostic criteria (aged 36-60 years, average 47.1) and 30 control subjects (aged 35-57 years, average 46.2). The study protocol was approved by the Medical School Ethics Committee, Zagreb, Croatia. All subjects completed a general health and nutritional questionnaire, and gave their written consent. Erythrocytes were hemolyzed and membranes were extracted by chloroform:methanol mixtures in order to obtain total lipids. After methanolysis, fatty acid composition was revealed by gas chromatography- mass spectrometry and spectrophotometric method was used for cholesterol measurement. RESULTS: In both groups of subjects the most abundant is the saturated palmitic acid (16:0) with 21.15% in control (C) and 22.05 % in PTSD group. The most abundant unsaturated fatty acid is oleic acid (18:1n-9) with 17.22% (C) and 20.45% (PTSD), followed by linoleic acid (C 18:2n-6) with 15.89% (C) and 15.62% (PTSD). Statistically significant differences were observed for a greater number of unsaturated fatty acids. Additionally, by comparison of saturated to unsaturated fatty acid ratio it has been confirmed that there is a higher percentage of saturated fatty acids in PTSD group (75%) than in the control group (72%). Moreover, a statistically significant difference (P≤0.005) has been established between the groups in n-3/n-6 ratio of PUFAs (0.030 C ; 0.015 PTSD). Statistically significant lower cholesterol concentrations were determined for the PTSD group (0.016 ± 0.01 mg/mL) than for the control (0.022 ± 0.01 mg/mL). The results from nutritional questionnaire revealed the fact that subjects from PTSD group consume fish more frequently. However, a greater number of overweight individuals was determined in both groups. CONCLUSION:Even though subjects from PTSD group consume fish as a rich source of essential n-3 PUFAs more often than the control group, they have twice as smaller ratio of n-3/n-6 PUFAs compared to the control group, which indicates to possible changes of lipid and fatty acid metabolism in people with PTSD.
