Sažetak
It has been postulated that colorectal cancer is caused or promoted by enviromental factors and especially by dietary factors that affect the enteric milieu. There are a variety of risk factors for colorectal cancer (general, genetic syndromes, familial and other diseases). The current data suggest that the development of colorectal cancer is a multistep process that involves a successive loss of genes. The most important independent pathologic factor for survival or recurrence following surgery is stage of the tumor. It became obvious that cellular immunity is the most important way of host defence against tumor growth. Two distinct mechanisms of cytotoxicity are involved in process of tumor growth: perforin and granzyme-dependent and Fas ligand-dependent mechanisms. Cytotoxic lymphocytes or killer cells comprise cytotoxic T lymphocytes (CTL) and natural killer (NK) cells. Both these subpopulations contain cytoplasmic granules with cytolitic molecule perforin The aim of our study was to compare the phenotypic profile of peripheral blood mononuclear cells (PBMNC) of patients with colorectal carcinoma with healthy people. We analysed the cytolitic activity of lymphocytes in peripheral blood of colon carcinoma patients classified as Dukes B or C, treated with three to four cycles of adjuvant chemotherapy with 5-Fluorouracil and Leucovorin. Colon carcinoma patients have significantlly higher amount of total perforin, double positive CD3+ P+ cells, and CD8+P+ cells, whereas the percentage of perforin positive cells in CD16 and CD 56 lymphocyte subsets are decreased compared to controls, pointing to the important role of cytolytic molecule perforin in immune reaction on aberrant cells.
